激光共聚焦显微镜成像原理及应用培训通知 

  研究所内一台转盘共聚焦显微镜 Olympus SpinSR10)以及一台高内涵细胞成像分析系统(Olympus FV-1200),将安排EVIDENT技术应用专家于20231031上午入所做相关理论培训。 

  Olympus SpinSR10 采用新型的Nipkow系统,可实现多色长时间超高分辨(120 nm)成像,成像速度快(200 fps),光毒性极低,适合长时间活细胞成像。本套设备配置了两个量子效率峰值高达95%的背照式sCMOS相机,配备了405 nm, 488 nm, 561 nm, 640 nm四根全固体长寿命激光器。搭载了硬件超分辨模块,搭配专利的软件超高分辨算法及100 NA 1.5的万能平场复消色差高分辨油浸物镜,共同实现转盘超高功能。 

  Olympus FV-1200配置405 nm488 nm559 nm635 nm激光器,可进行固定或活组织和细胞的微分干涉及荧光的断层扫描,多重荧光的断层扫描及重叠,实时动态扫描及其动态构建,荧光光谱分析,荧光各项指标定量分析,组织与细胞的三维结构动态构建,荧光共振能量转移(FRET)的分析,光漂白后的荧光修复(FRAP)等。

  使用转盘共聚焦显微镜和高内涵细胞成像分析系统的实验人员必须依次参加理论培训、上机操作培训(申请上机培训者必须参加理论培训)、带教,并通过考核,签订安全操作承诺书后,获得上机权限,目前使用线下登记双签制度。设备使用,采取预约登记的方式。 

  一、报名范围及培训要求 

  1.  理论培训不限范围,可到场签到,集中培训; 

  2.  上机培训限12人,每个课题组一个名额,同组内先报先得。

Training Notice for Principle and Application of Laser Confocal Microscope Systems 

There are a set of Spinning Disk Microscope System (Olympus SpinSR10) and a set of Laser-scanning Confocal Microscope System (Olympus FV-1200) of Analytical Core Facility. We will arrange a Chinese theoretical course and hands-on sessions by EVIDENT technical application on 31st Oct. 2023.  

Olympus SpinSR10 adopts a new Nipkow double turntable system, which can realize multi-color long-time ultra-high resolution (120 nm) imaging, fast imaging speed (200 fps), and extremely low phototoxicity, suitable for long-term living cell imaging. This set of equipment is equipped with two back illuminated CMOS cameras with quantum efficiency up to 95%, and four all solid-state long-life lasers of 405 nm, 488 nm, 561 nm and 640 nm are equipped. It is equipped with hardware super-resolution module, patented software ultra-high-resolution algorithm and 100 NA 1.5 universal flat fields achromatic high-resolution oil immersed objective lens to achieve the turntable ultra-high function. 

Olympus FV-1200 is equipped with 405 nm, 488 nm, 559 nm, and 635 nm lasers. It can perform differential interference and fluorescence tomography of fixed or living tissues and cells, multi fluorescence tomography and overlapping, real-time dynamic scanning and dynamic construction, fluorescence spectrum analysis, quantitative analysis of various fluorescence indexes, dynamic construction of three-dimensional structure of tissue and cell, and fluorescence resonance energy Quantitative transfer (FRET) analysis, photobleaching fluorescence repair (FRAP) and so on.

Experimental personnel using Olympus SpinSR10 and Olympus FV-1200 must participate in theoretical training, hands-on operation training (applicants for hands-on training must participate in theoretical training), mentoring, and passing the assessment. After signing a safety operation commitment letter, you can obtain hands-on permission. At present, the equipment adopts an appointment registration method and uses an offline registration double signature system. 

1. Application scope 

1) Theoretical course is not limited, and you can sign up offline; 

2) Only 12 persons can take part in the hands-on sessions. One quota for each group.  

2. Theoretical training arrangements: 

1) Theoretical course in Chinese: Principle of confocal microscope and the application of infection and immunity research.

2) Theoretical course in Chinese: New generation precise quantitative confocal microscopy imaging technology and application.  

Presented by EVIDENT Application Dr. Zhenhuan YUAN.

3. Hands-on Sessions in Chinese: 

1) Time: 2023/10/31 2:00-5:00pm, location: A324, Olympus FV1200 Laser Scanning Confocal Microscope Training (2 groups, each group 1.5 hour); 

2) Time: 2023/10/31 2:00-5:00pm, location: A326, Olympus SpinSR10 Spinning Disk Confocal Micorscope Training (2 groups, each group 1.5 hour).
