空间原位单细胞多组学技术进展讲座
空间组学被广泛誉为生命科学领域继单细胞测序之后一个新的前沿领域。单细胞空间多组学技术的出现,弥补了传统组学研究方法在空间和时间分辨率上的不足。它能够在细胞水平上揭示生物分子的异质性,为理解生物系统的功能和调控机制提供了更全面的视角。同时,该技术还有助于发现新的细胞类型和细胞状态,以及揭示细胞间的相互作用和通讯机制。CosMx 空间分子成像技术是NanoString(现Bruker Spatial Biology)于2022年底推出的空间原位单细胞成像技术,目前已经可以实现单细胞/亚细胞分辨率的6000plex以上RNA或70+蛋白的原位检测分析。
CosMx技术在神经科学、肿瘤学、发育生物学等领域取得了显著的应用成果。例如,在神经科学领域,该技术能够揭示神经元类型的空间分布和连接模式;在肿瘤学领域,可以研究肿瘤细胞的异质性和肿瘤微环境;在发育生物学领域,能够揭示细胞命运决定和胚胎发育过程。

Lecture on Advances in Spatial In Situ Single-Cell Multiomics Technology
Space omics is widely regarded as a new frontier field in the field of life sciences, following single-cell sequencing. The emergence of single-cell spatial multi omics technology has made up for the shortcomings of traditional omics research methods in terms of spatial and temporal resolution. It can reveal the heterogeneity of biomolecules at the cellular level, providing a more comprehensive perspective for understanding the functions and regulatory mechanisms of biological systems. At the same time, this technology also helps to discover new cell types and states, as well as reveal the interactions and communication mechanisms between cells. CosMx spatial molecular imaging technology is a spatial in situ single-cell imaging technology launched by NanoString (now Bruker Spatial Biology) at the end of 2022. Currently, it can achieve in situ detection and analysis of RNA or 70+proteins with a resolution of over 6000 plex at single-cell/subcellular level.
CosMx technology has achieved significant application results in fields such as neuroscience, oncology, and developmental biology. For example, in the field of neuroscience, this technology can reveal the spatial distribution and connectivity patterns of neuron types; In the field of oncology, the heterogeneity of tumor cells and the tumor microenvironment can be studied; In the field of developmental biology, it can reveal the determination of cell fate and the process of embryonic development.
